There are a number of experts on this site including "the one who never sleeps" goes by the name of Murray, who can help you but a little more information would be helpful. Are you speaking of modular or manufactured homes? When you say what are the differences the most obvious is one built on site one off site.
If you are speaking of modular you ca go step by step in the building process and find the difference but you have to know what you are looking for and without knowing who the builder or the manufacturer is that is impossible to do.
One of the differences is with a site built home you have more flexability in design but regardless which way you go choose your builder carefully.

As mentioned in the first reply, it depends on weather you are speaking of a Manufactured or truly a Modular home. First, if you are speaking of a Manufactured home, built to the HUD code, part of the difference will depend on where you live.
Different states have different building codes for housing so, in the case of a HUD coded home, in various states the differences will change.
After that, depending on the manufacturer and model then, you get to the "devil is in the details stage".
If you are speaking of a truly Modular home, it is built to the same building codes as the site built. Depending on the manufacturer and local builder/installer you can match just about anything a site builder can do, and match the quality of materials also. Usually you can to the matching at maybe up to a 1/3 less cost.
All three can be great housing choices depending on a couple of factors, how much money is in your budget and who is selling the home and building or installing it.
good luck
